In a futuristic, dark Budapest, there is a secret war between vampire and Lycans (werewolves) that has been going on for centuries. Selene (Kate Beckinsale) is a vampire warrior who accidentaly falls in love with an American played by Scott Speedman, who is bitten by a werewolf. This union is forbidden by their cultures, and causes a breakdown on both sides. Selene and Michael must fight for their love and their own survival.

Unbeknownst to humans, a battle has been raging for centuries between the unkempt, sewer dwelling Lycans and the fair-skinned, elegant Vampires. Selene (Kate Beckinsale) is a rubber clad Death Dealer; her duty is to hunt down and slay the Lycans one-by-one. It is on one such mission when Selene spies Michael (Scott Speedman) who is being tailed for mysterious reasons by a pack of Lycans. All of Selene's gun-slinging can't stop Michael from being bitten leading to a Shakespearean tragedy when the two find themselves having forbidden feelings for one another.

Set in present-day Europe, a war rages. Vampyres and lycans (werewolves) have never been able to co-exist since the day elder vampyre Viktor sentenced his own daughter to death for falling in love with and becoming pregnant by a lycan. Now an elder lycan has managed to find a way to join the two species together, half vampyre, half lycan, but stronger than both- by obtaining blood from a specific human. Selene, a vampyre determined to avenge her family's bloody death by lycans, vows to stop this blending of species until she meets Michael, and the truth about her past is revealed.

A war between vampires and Liken (werewolves) is going on. Selene, the main character, is one of the main vampires hunting the Liken. Lucien, one of the most powerful Liken, has been presumed dead by the vampires. Selene however has been finding evidence that Lucien may not be, but another vampire, who has a big ego and an even bigger crush on Selene - not shared - tells her not to look into it. Selene also finds that for some reason the Likens are stalking a human, and she wonders why, since that's not normal Liken behavior. She finds the human they're stalking named Michael, but he gets bitten by Lucien. She takes him to the vampire home mansion, where the other vampires find out about him and order Selene to stay in the complex. When Michael escapes, however, she follows him after waking the vampire that turned her, Viktor, from his hundred-year sleep, an act she gets into even deeper trouble for, desperate to learn why Lucien is still alive and what the Liken are doing with Michael.
